Julian Marsh (Jake Hoffman), an unemployed young lothario, is forced by his doctor dad (Chip Zien) to accept a job directing an off-Broadway play called Rosencrantz and Guildenstern Are Undead, which is described as a weird adaptation of Hamlet. The play has been written by a mysterious, pallid Romanian named Theo Horace (John Ventimiglia), a vampire who has just killed a young woman (Bijou Phillips). Unaware of the danger that surrounds Theo’s play, Julian casts his best friend Vince (Kris Lemche) as Hamlet and uses his new job to impress his ex-girlfriend, Anna (Devon Aoki), an aspiring actress. To Julian’s dismay, Anna has taken up with a shady businessman named Bobby Bianchi (Ralph Macchio), who has reputed ties to the Mafia. When Anna is cast as Ophelia, she slowly gets involved with Theo. The roles of Rosencrantz and Guildenstern in Theo’s play are filled by Carlo (Carlos Velazquez) and Mickey (Mike Landry), two narcissistic actors as foolish as the characters they are cast to play. Opening night draws near and people begin to die. Two bumbling New York City detectives (Jeremy Sisto and Lou Carbonneau) chase suspects as the movie builds to its climactic sequence.

Starting with the birth of the Jesus child, the film deviates wildly from the New Testament template, with the Virgin Mary now giving birth to twins, likely to give credence to the idea that Gnostic The Gospel of Thomas was actually written by the twin brother of Jesus. However, in keeping with tradition, only the life and suffering of the One is portrayed. Throughout the entire film, Jesus is shadowed by dark agents (the Four Evangelists) who seem to be the puppet masters of the story. But this preordained life spins out of control at the Last Supper, where Jesus develops a fondness for drinking blood. On his way to Calvary, he meets Saint Veronica and surrenders to this thirst, thereby acquiring immortality as a vampire. Jesus lives.

Jacob (Doug Bradley) is an antiques dealer who sets off with his new young and pregnant wife Lauren (Grace Vallorani) and his stepdaughter, Rachel (Rita Ramnani) to a forest homestead in a hopeful effort to heal bonds between feuding family members. He also carries a large, mysterious and ancient Babylonian mirror, smuggled out of the middle east, which he plans to sell for profit. Lilith (Natalie Celino), a sultry ornithologist appeares out of a bird hide by a noisy pair of friends hiking through the woods. Gaining their confidence, she and one of the men, Travis (Scott North) go into the woods for a quick tryst, but immediately there is a blood curdling scream. The other camper, Stanley (James Fisher), rushes toward the sound and finds his friend mutilated and near dead. Near Travis, Lilith explains to Stanley that the shadows had come to life and attacked them, and the two set off to seek help. When they end up at Jacob's cabin, the entire group comes under attack by supernatural forces of darkness. Rachel discovers Irish cowboy Phelan (Jonnie Hurn) hanging out in their barn. Phelan is a vampire and himself a former vampire hunter. He tells Rachel that the real danger to everyone is Lilith, and explains how Lilith had been Adam's willful first wife who, since having been exiled from the Garden of Eden, had been spawning demonic offspring in the world of man. Considered the mother to the Succubi, ruler of shadows, slaughterer of children, and consort to the devil himself, she had been re-awakened by, and drawn to the location of, the occult power inherent in the mirror carried by Jacob. Phelan has been harboring a centuries-long grudge against her for she having "turned" him 120 years earlier, and he had been tracking her for decades, equipped with the one relic that might finally kill her... a bone splinter from the rib of Adam himself.

The pair experiment on students in the school basement hoping to discover the secret of reanimating corpses (akin to the work of Victor Frankenstein). Their hopes are answered when they discover a solution of Monami's blood holds the properties to bring life to dead body parts and inanimate objects.

I Pass for Human follows the downward spiral of Jane (Eleanor Whitledge) following the heroin overdose and death of her boyfriend, Dax (Bryan Small). Finding heroin alone inadequate, in short order she replaces Dax with the company of new beau Rick (Josh Coxx). Not only is Rick a heroin addict, but he also has a dead significant other, Azami, whose jealousy apparently follows him from beyond the grave. The film plays on the conceit that heroin addicts escape death; in I Pass for Human, heroin leaves its users never quite dead, yet never quite alive.
