Six ans après les événements du premier film (Les Mondes de Ralph), Ralph La Casse et Vanellope restent toujours les meilleurs amis du monde et s'amusent chaque nuit à jouer ensemble. Alors que Ralph est heureux de la vie qu'il a, Vanellope avoue être ennuyée par son jeu, qui est trop prévisible, et aimerait de la nouveauté. Un jour, Ralph essaie d'exaucer son souhait en créant une course bonus dans l'un des circuits. Vanellope va contre la joueuse de la borne d'arcade pour profiter de la course à fond, tant et si bien que la joueuse, en forçant trop sur le volant, le casse. La situation paraît désespérée lorsque le propriétaire de la salle d'arcade, M. Litwak, annonce que le créateur de Sugar Rush a fait faillite, quand l'un des adolescents trouve un volant sur eBay. Cependant, M. Litwak le trouve trop cher et n'a d'autre choix que de débrancher la borne de Sugar Rush, laissant tous les personnages du jeu sans abri.

In fictional Oakton City, purple squirrel Surly and his mute rat partner, Buddy, reside in Liberty Park where their thieving reputation has made them outcasts. A group of urban animals led by Raccoon and his cardinal assistant, are running low on food for winter. Red squirrel Andie and gray squirrel Grayson, a self-styled hero, compete with Surly and Buddy to scavenge from a peanut cart manned by criminals Lucky and Fingers, who are casing a bank. The squirrels' efforts inadvertently end with the cart's propane tank exploding in the park and destroying the tree, where the animals store their food. Surly is banished and Buddy goes with him.

The young orphan Markussi lives with his little sister Mipoulok in an Inuit clan. Markussi can talk to animals and has some special powers unique to shamans. He does not want to be a shaman though and mainly keeps his powers a secret because he fears being a shaman would make him like the clan's shaman Croolik. Croolik grew selfish and unjust after the deaths of his sons, who died along with Markussi's father in an accident on a hunting trip. Croolik blames his sons' deaths on his now-separated wife Saya (whom he accuses of having let their sons go hunting at too young an age) and Markussi's father; by extension, Croolik also hates Markussi.

In Scotland, Merida, a young princess of the clan Dunbroch, is given a bow and arrows by her father, King Fergus, for her birthday. Her mother, Queen Elinor, is dismayed. The film opens with Scorch Supernova (Brendan Fraser) on a mission to rescue captured babies from the Gnarlachs. He rescues them just in time before the Gnarlachs wake up. Scorch then returns to Planet Baab where he is known as a famous hero and a love interest for anchorwoman Gabby Babblebrook (Sofía Vergara) while at BASA his brother Gary (Rob Corddry) works. Gary's son Kip (Jonathan Morgan Heit) is a big fan of his uncle Scorch. Soon, Gary and Scorch receive a message from Lena Thackleman (Jessica Alba), the head of BASA, that Scorch will be sent to the "Dark Planet" (Earth) due to an SOS call. Scorch decides to go on the mission to the Dark Planet, but Gary strongly insists that he doesn't go because he is not serious. Also, no alien has ever returned from the dark planet. After further arguing, Gary finally says that he won't be helping Scorch and quits BASA before Scorch himself fires him. Gary then goes home to his wife Kira (Sarah Jessica Parker) and Kip only to find out that Scorch has already gone on the mission to the Dark Planet, while Kip is watching it on live TV in excitement.

Rango, a pet chameleon (Johnny Depp) becomes stranded in the Mojave Desert after his terrarium falls from his owners' car by accident. A bee named Barry B. Benson (Jerry Seinfeld) has recently graduated from college and is about to enter the hive's Honex Industries honey-making workforce alongside his best friend Adam Flayman (Matthew Broderick). Barry is initially excited to join the workforce, but his latent non-conformist attitude emerges upon discovering that his choice of job will never change once picked. Later, the two bees run into a group of Pollen Jocks, bees who collect pollen from flowers outside the hive. The Jocks offer to take Barry outside the hive to a flower patch, and he accepts. While on his first pollen-gathering expedition, Barry gets caught in the rain, and ends up on the balcony of a human named Vanessa (Renée Zellweger). Upon noticing Barry, Vanessa's boyfriend Ken (Patrick Warburton) attempts to swat him, but Vanessa instead safely catches and releases Barry outside the window, saving his life.
