Makabe Rokurota (Hiroshi Abe), a loyal retainer of Princess Yuki (Masami Nagasawa), has been commissioned to transport Yuki and Akizuki's ample treasury of gold bars safely to the politically stable Hayakawa. The story follows the escape of two men from an Alaska prison, the efforts of a railroad dispatch office to safely stop the out-of-control train they are on, and the hunt by their warden to recapture them. Oscar "Manny" Manheim is a ruthless bank robber and hero to the convicts of Stonehaven Maximum Security Prison. After two previous escape attempts the doors to Manny's cell have been welded shut for three years. A court order compels Manny's nemesis, the vindictive and sadistic Associate Warden Ranken, to release him back into the general prison population. Manny intends to break out a third time with his older brother Jonah Manheim, but is forced to set his escape plan into action in the middle of winter. Ranken employs a serial killer to give Manny the incentive, by stabbing him grievously through his left hand. Jonah fatally stabs the serial killer to death in retaliation, and in turn is severely beaten by the prison guard, leaving him in a high-security hospital wing.

In a distant galaxy lies the desert planet of Ura, which has two suns. There, two rival warlords, Zeg and Bal Caz, constantly fight against each other in a battle over the village's only wellspring. The mercenary warrior Kain emerges and announces that his skills are for hire to the highest bidder. Naja, a beautiful sorceress that has been taken captive by Zeg changes Kain's purpose to take the well for himself, instead choosing to save Naja and the village people. Kain starts entangling the situation, and by taking advantage of the ongoing feud, he seeks to debilitate the rival warlords and defeat them.

The film focuses on the lives of a variety of characters who happen to live in a rubbish dump. The first to be introduced is a mentally challenged boy who lives in a world of fantasy in which he is a tram conductor. He is both the tram and the tram driver and follows a set route and schedule through the dump; his dedication to the fantasy is fanatical. The film title refers to a Japanese onomatopoeia for the sound made by a tram or train while in motion ( "Do-desu-ka-den do-desu-ka-den do-desu-ka-den"). The sound is made by the boy as he makes his daily faux-tram route through the dump.

The film takes place in Koishikawa, a district of Edo (the former name of the city of Tokyo), in the 19th century. Young Dr. Noboru Yasumoto (Yūzō Kayama) is the film's protagonist. Trained in a Dutch medical school in Nagasaki, the arrogant Yasumoto aspires to the status of personal physician of the Shogunate, a position currently held by a close relative; his father is already a well-established, highly competent physician. Yasumoto believes that he should progress through the safe, and well-protected, army structure of medical education. However, for Yasumoto's post-graduate medical training, he has been assigned to a rural clinic under the guidance of Akahige ("Red Beard"), Dr. Kyojō Niide (played by Toshiro Mifune). Dr. Niide may seem like a tyrannical task master, but in reality he is a compassionate clinic director. Initially, Yasumoto is livid at his posting, believing that he has little to gain from working under Akahige. Dr. Yasumoto feels that Dr. Niide is only interested in his medical notes and soon rebels against the clinic director. He refuses to wear his uniform, disdains the food and spartan environment, and enters the forbidden garden where he meets "The Mantis" (Kyōko Kagawa), a mysterious patient that only Dr. Niide can treat.

A wealthy executive named Kingo Gondo (Toshiro Mifune) is in a struggle to gain control of a company called National Shoes. One faction wants the company to make cheap, low quality shoes for the impulse market as opposed to the sturdy but unfashionable shoes currently being produced. Nine young samurai, including his nephew, believe that the lord chamberlain, Mutsuta, is corrupt after he tore up their petition against organised crime and said maybe he himself was behind the crimes. One of them tells the superintendent of this and he agrees to intervene. As the nine meet secretly at a shrine and discuss their problem, a ronin (Mifune) emerges from another room where he has been resting. The ronin has overheard their plans, and suggests that the superintendent is in fact the real corrupt official. While at first the samurai are insulted by his claims, they soon find themselves surrounded by the superintendent's men, proving that he was correct. The ronin hits a few of the attackers and bluffs their leader into leaving, taking only a small amount of money in reward to buy food because he is starving; however, after realizing that Mutsuta and his family must now be in danger, he decides to help the samurai bring down the corrupt officials.

The film begins with a group of news reporters watching, and gossiping, at an elaborate wedding reception for Yoshiko Iwabuchi (Kyoko Kagawa), the daughter of Vice President Iwabuchi (Masayuki Mori, the villain character) of the Unexploited Land Development Corporation, a construction company, and Koichi Nishi, the president's secretary (a bespectacled Toshiro Mifune). The police interrupt the wedding when corporate assistant officer Wada, who is the wedding reception's master of ceremony, is arrested on charges of bribery in a kickback scheme. The reporters comment this incident is similar to an earlier scandal involving Iwabuchi, administrative officer Moriyama, and contract officer Shirai. That earlier case was hushed up after the apparent suicide of Assistant Chief Furuya, by jumping off the corporate office building, creating a dead end in the investigation before any of the company's higher-ups could be implicated.

A Mexican village is periodically raided for food and supplies by Calvera (Eli Wallach) and his bandits. As he and his men ride away from their latest visit, Calvera promises to return to loot the village again. Taking what meager goods they have, the village leaders ride to a town just inside the American border hoping to barter for weapons to defend themselves. While there, they encounter Chris Adams (Yul Brynner), a veteran Cajun gunslinger. After listening to their tale, Chris suggests that the village hire gunfighters, which would be cheaper than guns and ammunition. The village men relentlessly try to convince him to be their gunman. At first he agrees only to help them find men, but later he decides to recruit six other men to help him defend the village, despite the poor pay offered.
