A humanoid alien (Aamir Khan) lands on Earth naked on a research mission in Rajasthan, but is stranded when the remote control of his spaceship is stolen, though he manages to get the thief's cassette recorder, a National Panasonic RQ-565D. On the same day in Bruges, Jaggu (Anushka Sharma) meets a man named Sarfaraz (Sushant Singh Rajput) and falls in love with him. Jaggu's father (Parikshit Sahni) objects to their relationship because Sarfaraz is a Pakistani Muslim, while Jaggu is a Hindu. He consults godman Tapasvi Maharaj (Saurabh Shukla) who predicts that Sarfaraz will betray Jaggu. Determined to prove them wrong, Jaggu proposes to Sarfaraz. She is heart-broken at the wedding chapel when she receives a letter calling off the marriage due to their cultural differences.

This movie is about three childhood friends, Akash Malhotra (Aamir Khan), Sameer Mulchandani (Saif Ali Khan), and Siddharth "Sid" Sinha (Akshaye Khanna) who, after college, fall in love. The story revolves around three roommates, journalist Tashi (Imran Khan), photographer Nitin Berry (Kunaal Roy Kapur) and cartoonist Arup (Vir Das), leading an unkempt and debt-ridden life in a shoddy apartment. Tashi's ditzy fiancée, Sonia (Shenaz Treasurywala), is an air hostess who agrees to deliver a package for Vladimir Dragunsky (Kim Bodnia) to Somayajulu (Vijay Raaz), without realizing its contents or that Somayajulu is a gangster. Sonia asks Tashi to deliver the package. Tashi, in turn, asks Nitin to do so. But Nitin is unable to do so as he is suffering 'Delhi Belly'. Nitin hands Sonia's package to Arup for delivery to Somayajulu, along with a package containing his stool sample for delivery to Nitin's doctor. Arup mixes up the two bags. Somayajulu, furious, tortures Vladimir to find his package.

Natha Das ManikPuri (Omkar Das Manikpuri), better known as Natha, is a poor farmer from the village of Peepli in "Mukhya Pradesh", who struggles to farm enough money for his family made up of his elder brother, Budhia Das Manikpuri (Raghubir Yadav), his wife, Dhaniya (Shalini Vatsa) and his ailing mother, Amma (Farookh Zafar), who spends most of her time lying down and screaming at Natha and Dhaniya. Natha and Budhia often pretend to go out farming when actually they save up whatever money they have to buy alcohol. This has left the whole family famished and now the banks are demanding repayment of loans or else the family will be stripped off their land and house. In the meantime, the Mukhya Pradesh Government have called a by-election due to continuous criticism of their blind eye towards the desperate poverty that surrounds India's largest state.
