La barrière protégeant les humains du royaume des démons étant tombée, des hordes de démons affluent dans le monde humain. Le roi des démons envoie alors le légendaire chasseur de démons Pu Songling (Jackie Chan), un homme de haute culture, intellectuel, sophistiqué et rusé, pour retrouver chacun d’eux. Équipé du pinceau de calligraphie magique du roi, il est aidé dans sa mission par un groupe hétéroclite de gentils monstres. Il fait équipe avec l'officier de police Fei (Austin Lin ) pour résoudre une affaire de disparition en série d'adolescentes dans un village. Il apprend bientôt que les filles ont été enlevées par la belle démone Nie Xiaoqian (Zhong Chuxi ) et un autre démon rusé (Lin Peng ). Tandis que Songling mène son enquête, il rencontre un autre chasseur de démons, Chixia (Ethan Juan ), en pleine bataille avec deux démons. Songling défait le démon rusé et sauve Chixia avant de l'emmener dans son atelier. Il apprend que Xiaoqian était autrefois un homme tombé amoureux du démon Caichen, un serpent au bon cœur. Caichen a donné à Xiaoqian une orbe spirituelle en échange de son ombre, mais l'orbe la transformée en démon. Au lieu de piéger les âmes, Songling doit maintenant en sauver une.

Élevé par son oncle Wu Sanxing (Wang Jingchun), archéologue renommé, Wu Xie (Luhan), diplômé d'une grande école d'architecture, est fasciné par l'architecture ancienne et les antiquités. In western China, 48 BC, a security company under China's current government called the Silk Road Protection Squad is committed towards stopping battles and promoting the idea of peace. Huo An (Chan), the captain, successfully averts a battle between the Indians and Huns. The group returns to their unnamed home city, where Huo An's wife serves as a schoolteacher for orphans. However, the government uncovers evidence that someone in the group is corrupt, causing the entire group to be sentenced to construction work at Goose Gate, a ruined fortress. Once they arrive, the group pays respects at the shrine of a fallen Chinese general, since they originally were child-slaves of Hun descent, but later the general had rescued them.

IDC (International Defense Commission) agent Sean Wong (Andy On) and his team members are in Jordan on a dangerous mission to escort a scientist named Kenner Osama Muhammad, who has stolen a copy of the smallpox virus, to Norway. On the way out of the country, the convoy were ambushed by a group of assailants. It was later revealed that Sean has betrayed the team for his own agenda and shot his colleagues Jon Man (Jay Chou) and his girlfriend Ice (Michelle Bai). Sean then escapes with Kenner and the virus. The failed mission left Ice dead while Jon has a bullet lodged in his brain with no safe way to remove it and only two weeks to live. Three months later, the crew of a freight ship on Malaysian water was found to have been infected and killed by the smallpox virus. Elsewhere, Sean contacts an arms dealer called Tyler (Jared Robinson), who suggests to mutate the virus into a biological weapon, develop a vaccine and sell it to a corrupt pharmaceutical company. In Beijing, Jon returns home to spend his remaining days with his mother (Elaine Jin), who tells him that he has a long lost brother, Man Yeung (Nicholas Tse) whom she left behind with his father, Man Tin (Liu Kai Chi).

Yan Chixia's master sent him to Black Hill to train in the arts of demon hunting. After capturing an attractive female demon, Nie Xiaoqian, Yan falls in love with her and realises that his master sent him there to make him learn how to control his emotions. Xiaoqian also falls in love with Yan, and while he feeds her candy, she says she will fall in love with anyone who fed her candy. After some time, she begs him to kill her because humans and demons are not supposed to be together. Yan stabs her with his magic dagger, which does not kill Xiaoqian, but causes her to lose her memory of their relationship. Yan arrives late for a battle against the Tree Demon, Laolao, and sees that several of his fellow demon hunters have already fallen. The last one, Xia Xuefenglei, embeds his arm into Laolao and tells Yan to chop it off, thereby banishing Laolao from the mortal world.
