Lobo is based on the life of Mikel Lejarza, an agent of the Spanish intelligence service in the 1970s. During the last stages of Franco's dictatorship between 1973 and 1975, Lejarza infiltrated ETA, a paramilitary group seeking independence for the Basque Country, and was responsible for the downfall of one quarter of the terrorist activists of the organisation - including Special Forces members and some top-level figures of the group.

Yolanda, a cabaret singer, brings heroin to her lover who drops dead of an overdose. To escape from the police, the singer looks for refuge in a local convent. The Mother Superior, a fan of Yolanda, rapturously greets her. The mission of the order, called the Humiliated Redeemers (Redentoras humilladas), is to offer shelter and redemption to fallen women. The convent once was a bustling haven for prostitutes, drug addicts and murderers, but it is now in disrepair. The order is facing serious financial hardships as their prime financial supporter, the vain and greedy Marchioness (La Marquesa) has decided to discontinue the convent's annuity under the pretense of economizing. She feels less affinity with the sisters than did her recently deceased husband, who set up a fund in gratitude. The convent had taken in their wayward daughter, Virginia, who subsequently became a nun and ran off to Africa, only to be eaten by cannibals.

Paul Marnac (Paul Naschy) and his infirm wife Geneviève (Julia Saly) move to his childhood estate in the French countryside. The estate is also the site of the castle inhabited by Paul's ancestor Alaric de Marnac, who was known for brutally slaughtering anyone whom he suspected of infidelity. The American television news reporter Dean Miller (Hugo Stiglitz) waits at a European airport to interview a scientist about a recent nuclear accident. An unmarked military plane makes an emergency landing. The plane doors open and dozens of deformed people burst out and begin stabbing and shooting the military personnel outside; they prove to be resistant to the most grievous wounds and are relentless in their assault, stopping only to consume the blood of their victims. Miller tries to alert the public, but General Murchison of Civil Defense (Mel Ferrer) will not allow it. Miller tries to find his wife Anna (Laura Trotter), who works at a hospital, as the crazed assailants overrun the city, their ranks swollen by many of their former victims.
