After having killed the notorious extortionist Mayil Vaaganam in the first film, Duraisingam (Suriya) has gone undercover after meeting the Home Minister and is working as an NCC officer in a school in Thoothukudi. The only people who know about this operation are the Chief Minister and Home Minister apart from himself. Sathya (Hansika Motwani) is a student who falls for Duraisingam though he is waiting to marry Kavya (Anushka Shetty), his love interest from the previous film. Susai (Santhanam), a local who works in the same school shows Duraisingam parts of Thoothukudi and also tells him about the rogue elements dominating the area helping him in his surveillance. He requests the Home Minister to transfer Erimalai (Vivek) and another cop for assisting him. Sathya's moves are frequently repulsed by Duraisingam who learns that Sathya is the niece of Thangaraj (Rahman). Bhai (Mukesh Rishi) and Thangaraj, are two criminals who control the area. Bhai and Thangaraj have connections with an international gangster and drug lord Danny (Danny Sapani).

The film begins in Nallur, a small village in Thoothukudi district in southern Tamil Nadu. Duraisingam (Suriya), is the honest police sub inspector of Nallur, assisted by his bumbling colleague Erimalai (Vivek). He belongs to Nallur with his father having a respectable status in the village. His family business is provision stores and Duraisingam wants to join it, but he joined police due to his father's wishes. He resolves most of the problems in his village with non-violence and mutual counselling. He uses force only when the situation demands it, thereby gaining much reputation and love from the villagers. Mahalingam (Nassar) is an industrialist in Chennai and a friend of Singam’s father (Radha Ravi). He comes to the village with his beautiful daughter Kavya (Anushka Shetty). Eventually Singam and Kavya fall in love with each other.

When a young boy Saravanan is killed by rowdy - and aspiring politician - Dhanasekaran(Sayaji Shinde), his distraught mother(Vinaya Prasad) is ready to kill herself. But she is stopped by a man Mani Shankar(Vijayakumar) who promises her that her son will be reincarnated and meet her 30 years hence. Marudhamalai (Arjun), a police constable, goes to work in the village of Nachiapuram after passing his police exams and getting a merit. Here, he meets 'Encounter' Ekambaram (Vadivelu) who is his senior officer. He is asked to do all sorts of work for him and one day, Marudhamalai loses a convict on the way to the court due to Ekambaran's soft heart. Then, later in the market, after Ekambaram is humiliated by a beggar, Marudhamalai sees Maasi, the 'boss' of the city, killing a person who stood for a candidate in the elections. Then the election commissioner comes to the place and he discovers that elections have not been held for 16 years in the city due to Maasi's opposition.

In the South Indian kingdom of Cholapuram Paalayam in the year 1771, Raja Mokkaiyappar and his queen, Rajamatha Bhavani Ammaiyar are desperate for a child, as all their previous twenty-two children have died at birth. Unknown to them, Bhavani's brother, Sangilimayan, who is also the Rajaguru (Chief Minister), is responsible for the death of their children. Sangilimayan is shocked to see that his sister has given birth to twins. Before informing the king about the birth, he calls the palace astrologer, Chinnavadayaan, who predicts that the elder twin will be incapable of making decisions on his own while the younger one will be smarter. Sangilimayan orders the palace doctor, Kailasakaruppan, to kill the younger one, but Chinnavadayaan tells Sangilimayan that his actions would not be good for the kingdom. Instead, Sangilimayan orders the palace doctor to abandon the child in a nearby river. The elder child is named Pulikecei XXIII. Maragathavalli, Kailasakaruppan's childless wife, rescues the abandoned child from the river, and the couple decides to raise him as their own, naming him Ukraputhan.

Saravanan (Rajinikanth), a psychiatrist, visits India on vacation. He meets up with his foster brother Senthilnathan, alias Senthil (Prabhu), and his wife Ganga (Jyothika). The film opens with a flashback showing the family feud between families of Prabhakaran (Vijayakanth) and Bhavani (Indraja). Prabhakaran and Bhavani are in love. The murder of Bhavani's father, is wrongly accused on SP's father. He starts to help Bhavani's family and mortgages his own house to finance her factory with the intention of making it up to them as he still believes that it's his father who killed Bhavani's father. But Bhavani is now trying to take revenge for her father's murder. SP's mother is now on her deathbed. While in the hospital, his mom tells how his father had a different wife and he should ask them forgiveness. He storms into Bhavani's house and accuses her uncle (who really killed Bhavni's dad), also trying to make her understand the truth but she doesn't believe him. Then Prabhakaran says he doesn't love her any more and vows that he will never trust a woman by remaining a chronic bachelor forever.
