While carrying out his research as a travel journalist, cynical American Pete McKell (Michael Vartan) joins a group of tourists on a crocodile watching river cruise in Kakadu National Park of Australia's Northern Territory, led by wildlife researcher Kate Ryan (Radha Mitchell). In Burundi, a British forensic anthropologist is examining the corpses in a mass grave, claiming they were all killed in an identical manner. When the woman digs her shovel into what she believes is another grave, an unseen creature attacks, and violently drags her into the river. Tom Newman, a hydro-electrical engineer, arrives in Africa with his family to help build a dam. His daughter, Jessica, isn't getting along with her stepmother, Amy, because she is not happy about her parents divorce. The next morning, Amy, Jessica and her brother David go on a game drive with Brian, a ranger, while Tom goes to the dam. While driving off-road, David asks the ranger to stop the jeep because he has to defecate. Brian grabs his rifle and escorts David to a nearby tree. They encounter two lionesses, backed by a lion behind them. One lioness attacks and David runs back to the car, where Brian sacrifices himself to save the others. The keys lost, Amy, Jessica, and David are trapped in the car and stalked by the lions, who devour Brian. Back at the lodge, Tom is informed of his family's disappearance, and attempts to contact Crawford, a professional hunter and guide, at the suggestion of the park rangers. Crawford refuses to help him, as he is a big five hunt guide, not a leader of search parties. The next day, after not being able to accompany the rangers in the air, Tom goes to see Crawford, and manages to hire his services at the latter's own price. Back in the car, David spots Brian's keys, and Amy retrieves them, only to wreck the truck when she drives the wrong way. Crawford and Tom look for signs of them while checking in on the rangers trail periodically, but the rain the previous night has washed away any tracks. When the rangers fly by without noticing them, Jessica gets out and tries to draw their attention. She fails and, along with Amy, is attacked by one lioness, which is killed by two native hunters.

Anthropological professor Conrad Hamilton attempts to study a new species of primate, possibly the missing link between humanity and the great ape, found in a hidden valley deep within the jungles of Thailand. Hamilton's initial research team tries to capture one of these new (and very large) primates, but fail and are all killed. A great white shark follows a team of water skiers. The driver stalls the ski boat but manages to restart it before the shark can approach. The shark follows the water skiers into the SeaWorld park and throws the gate off its rails while it is closing. Meanwhile, Florida announces the opening of SeaWorld's new glass covered underwater tunnels that allow close up views of the park's animals.

After witnessing gangster Eddie Kim brutally beat U.S. Prosecutor Daniel Hayes to death in Hawaii, Sean Jones is escorted by FBI agents Neville Flynn and John Sanders on a Boeing 747-400 to testify in a trial in Los Angeles. Despite increased security for the flight, Kim arranges for a time-release crate full of venomous snakes to be placed in the cargo hold in an attempt to bring down the plane before it reaches Los Angeles International Airport (LAX). To ensure the snakes attack the passengers without being provoked, he has one of his henchmen disguised as an airport ground employee spray the passengers' leis with a special pheromone which makes the snakes highly aggressive. The crate opens midway through the flight and the snakes make their way through the cabin. A couple having sex in the bathroom, and a man using the bathroom are the first killed. The plane's captain, Sam McKeon, investigates and fixes an electrical short, but is killed by the viper that caused it. Co-pilot Rick, unaware of the snake, believes Sam has suffered a heart attack and continues toward LAX.

During the demonstration of a device which allows the observation of alternate dimensions, an accident causes the building in which the demonstration is taking place - and its occupants - to be transported to a dimension seemingly devoid of animal life but for large, vicious carnivores which hunt and kill the displaced humans one by one. A creature with six eyes attacks the people. The senator is attacked and ripped in half, and Lex has a heart attack. While researching an alien creature, Josh Burke accidentally sprays Al with an acidic toxin, and the first creature kills the private. While out getting water, two groups of three include Sam, Josh, and Dr. Karen Frost in one group, and Rivers, Dr. Jillian O'Hara and Murphy in the other, try to get out. Josh steals an egg, drops it on the rock, and is instantly sliced in half by "Momma." Rivers blows himself up to save Sam, and Brian Murphy is attacked and killed by "Momma". Only Colonel Synn, Dr. Frost, and Dr. O'Hara are left. Dr. Frost goes off by herself in an attempt to find intelligent life because of a structure she saw in the distance, only to discover too late that it was part of a building that got beamed with them and she is attacked and eaten by "Momma" off-screen. At the same time Colonel Synn and Dr. O'Hara manage to repair the device and return home, but discover that upon their return, off-screen damage from the machine has apparently caused chaos in their world, presumably bringing through one of the monsters.
