Five years after saving New York City from the demi-god Gozer, the Ghostbusters—Egon Spengler, Ray Stantz, Peter Venkman, and Winston Zeddemore—have gone their separate ways after having been sued by the city for property damage and barred from investigating the supernatural, forcing them out of business. Ray owns an occult bookstore and works as an unpopular children's entertainer with Winston, Egon works in a laboratory on the Columbia University campus, conducting experiments into human emotion, and Peter hosts a pseudo-psychic television show. Peter's former girlfriend Dana Barrett has had a son, Oscar, with a violinist that she married then divorced when he received an offer to join the London Symphony Orchestra. In order to take care of her baby, Dana quit her former cellist profession and now works as a restorer at the fictional Manhattan Museum of Art, working to prepare a malevolent-looking portrait of a legendary sixteenth-century tyrant named Vigo the Carpathian of Moldavia for an upcoming exhibition.

A group of corpses are found in a tunnel, where they were attacked and killed by Chupacabras. A freak cyclone hits Los Angeles causing man-eating sharks to be scooped up in water spouts and flood the city with shark-infested seawater. Bar-owner and surfer Fin sets out with his friends, Baz, Nova, and George, to rescue his estranged wife, April, and their teenage daughter, Claudia, after the bar and boardwalk is destroyed in flooding. While heading to April's home, the group stops in a freeway to save people as flooding causes sharks to attack. George is killed and the group learns of a tornado warning. They arrive at April's house just before the first floor is flooded and shark-infested. Collin, April's boyfriend, is eaten by sharks, but the rest of the group escape unharmed.

Hiro Hamada is a 14-year-old robotics genius in the futuristic fictional city of San Fransokyo. Raised by his aunt Cass and older brother Tadashi after the death of his parents, he spends his time participating in illegal robot fights. To redirect Hiro, Tadashi takes him to the robotics center at his university, where Hiro meets Tadashi's friends: GoGo, Wasabi, Honey Lemon, and Fred, as well as Baymax, the inflatable healthcare companion robot Tadashi created. To enroll in the school, Hiro signs up for the school's science fair and presents microbots: swarms of tiny robots that can link together in any arrangement imaginable. Professor Callaghan, the head of the university's robotics program, is impressed, and grants Hiro entrance to the university. Alastair Krei, renowned entrepreneur and president of Krei Tech, offers to buy the microbots, but Hiro follows Callaghan's warnings about Krei's questionable business practices and declines to sell them. When Tadashi and Hiro later arrive at the university to find it engulfed in flames, Tadashi rushes in to rescue Callaghan, but the building explodes moments later, killing him.
