Loosely based on the video game of the same name, the film centers on the character of Rayne, an unholy breed of human and vampire called a Dhampir. Dhampir are unaffected by crucifixes and do not thirst for human blood. She is the daughter of the Vampire King Kagan who has gathered an army of thralls, both vampire and human, in order to annihilate the human race. She was conceived when Kagan raped her mother, and she later witnessed him killing her.

In 1818, Abraham Lincoln (Benjamin Walker) lives in Indiana with his parents, Nancy (Robin McLeavy) and Thomas (Joseph Mawle), who works at a plantation owned by Jack Barts (Marton Csokas). There, Lincoln intervenes when he sees his friend, a young African American boy, William Johnson (Anthony Mackie) being beaten by a slaver. Because of his son's actions, Thomas is fired. That night, Lincoln sees Barts break into his house and attack his mother. Nancy falls ill the following day, and dies shortly afterwards. Thomas tells Lincoln that Barts poisoned her, but asks that he promise not try to avenge her death.

The prequel is set in Mexico in the early 1900s and begins with an American author, Ambrose Bierce, experiencing a nightmare in which he dies at the hands of Pancho Villa. Bierce wakes and talks to a local bartender about his intentions to join Pancho Villa's revolutionary army. He joins a stagecoach transporting a newly-wed couple, John and Mary Newlie, who are traveling to Mexico to preach Christianity. Meanwhile, Johnny Madrid, a dangerous local outlaw, escapes from the gallows and kidnaps his hangman's beautiful daughter, Esmeralda. Madrid receives assistance from Reece, a young woman who wants to become Madrid's apprentice as an outlaw. With the hangman and a local posse on their trail, Madrid meets with his gang. They later rob Bierce's stagecoach because of Reece's belief that Bierce possesses an invaluable object. The object turns out to be the manuscript for Bierce's new book. Annoyed by this, Madrid leaves Reece to die in the desert. She is found by the posse who use her to track the two.

Barrow, Alaska is preparing for its annual "30 Days of Night", a period during the winter when there is a month-long polar night. As the town gets ready, a stranger rows ashore from a large ship. Reporter Sadie Blake has just published a notable article featuring a secret Gothic party scene. The very night following the publication, one of Sadie's sources, Tricia Rawlins, is invited by her friend Kaitlyn to an isolated house in which such a party is to take place. Tricia is reluctant to enter with the curfew set by her strict father, so Kaitlyn goes alone. When she does not return, Tricia becomes worried and enters the house as well. To her horror, she finds Kaitlyn in the basement with two vampires hanging onto her and drinking her blood. She tries to hide, but the vampires find her quickly. The next day, Sadie learns of the girl's death and decides to investigate the matter. She soon attracts the interest of the vampire cult, who kidnap her and bring her in for questioning.
