The film opens to scenes of Berlin during World War Two, with the ongoing war depicted by bombs and explosions, both onscreen and in the background soundtrack. The film then jumps back twenty years in time, and, through a series of vignettes about worker Hans Behnke, traces the way in which a typical worker who opposes Nazi party ideology could be drawn into complying and cooperating with the Nazi regime. Significant vignettes include depictions of the high unemployment in 1920s Germany and later, the threat to Hans's job due to his failure to belong to the Nazi party. It is implied that Hans's complicity with the Nazi regime rises out of a desire to be able to provide for his family and not return to the ranks of the Arbeitslos (unemployed).

The infamous Nazi doctor Josef Mengele, who performed unethical medical experiments and is considered to be personally responsible for the selection of mass groups of detainees to be murdered in the gas chambers at the Auschwitz concentration camp, comes back from his hideout in Argentina as an 87-year-old man who is in his last days. It is the middle of World War II and in the deserts of Africa, Sgt. Sullivan puts together a group of Italian-Americans into disguise as Italian soldiers in order to infiltrate a North African camp held by the Italians. After the soldiers have killed the Italians in their beds, they find a hooker living at the camp. Sullivan's commandos are to hold this camp and its weaponry until an American battalion arrives, all the while these Italian-Americans pretend to be Italian soldiers, often hosting the enemy. Cpt. Valli is a young, by-the-book officer who constantly argues with Sgt. Sullivan, who tells his superior that he has no idea what he is doing. One man on the base is an entomologist who is killed and then things get terrible.

The movie starts off with a man, named Schlomo (Lionel Abelanski), running crazily through a forest, with his voice playing in the background, saying that he has seen the horror of the Nazis in a nearby town, and he must tell the others. Once he gets into town, he informs the rabbi, and together they run through the town and once they have got enough people together, they hold a town meeting. At first, many of the men do not believe the horrors they are being told, and many criticize Schlomo, for he is the town lunatic, and who could possibly believe him? But the rabbi believes him, and then they try to tackle the problem of the coming terrors. Amidst the pondering and the arguing, Schlomo suggests that they build a train, so they can escape by deporting themselves. Some of their members pretend to be Nazis in order to ostensibly transport them to a concentration camp, when in reality, they are going to Palestine via Russia. Thus the Train of Life is born.

The picaresque story follows its protagonist, Pasqualino (Giannini) who, as a dandy and small-time hood in Naples, to save the family honour, is sent to prison for killing a pimp (and then dissecting the victim and placing the body in suitcases) who had turned Pasqualino's sister into a prostitute. Convicted and sent to prison, Pasqualino succeeds in being transferred to a psychiatric ward. Desperate to get out, he volunteers for the Italian Army, but then somewhere in Germany he deserts with a comrade. They are captured and sent to a concentration camp. There, in a bid to save his own life, Pasqualino decides to survive by providing sexual favors to the obese and ugly female commandant (Stoler). His plan succeeds, except for the fact that he is then put in charge of the barracks as a kapo, and is obliged to select six men to be killed under the threat that if he doesn't do so, they will all be killed. Pasqualino ends up executing the soldier with whom he was captured and being responsible for the death of another fellow prisoner, a Spanish anarchist. At the war's end, upon his return to Naples, Pasqualino discovers that his seven sisters, his fiancée and even his mother have all survived through prostitution.
