Robert Mark Kamen is an American screenwriter who has been writing major motion pictures for over twenty-five years. He is best known as creator and co-creator of the Karate Kid and Transporter franchises, as well as the 2008 action thriller Taken.

Au début du film, l'agent du Secret Service Mike Banning combat violemment des soldats d'élite, dans un centre d'entraînement opéré par la société paramilitaire Salient Global, qui appartient à son ex-collègue des forces spéciales et ami Wade Jennings. Par ailleurs, Banning cache à son entourage qu'il souffre de migraines, d'insomnie et de maux de dos et consulte plusieurs médecins pour les traiter. Plus tard, il accompagne le Président des États-Unis, Alan Trumbull, à une partie de pêche sur une rivière. Sur le bateau de pêche, lorsque ses douleurs l'empêchent de poursuivre son travail, il demande à être remplacé par un collègue. Près de la rive, Banning aperçoit un essaim se diriger vers le site et comprend qu'il s'agit de drones. Les hommes sur place sont tués, sauf Banning et Trumbull.

Former covert operative Bryan Mills visits his daughter, Kim, who has just discovered she's pregnant, at her L.A. apartment to deliver an early birthday gift. At the funeral of his son Marko and associates in Tropojë, Albanian mafia head Murad Hoxha vows to seek vengeance on his son's killer. In 1992, Bogota, Colombia, Fabio Restrepo (Jesse Borrego) has key information about drug lord Don Luis Sandoval (Beto Benites). However, Don Luis fears that Fabio will be putting a hit on them, so he sends his henchman Marco (Jordi Mollà) to kill him. Fabio gives his nine-year-old daughter Cataleya (Amandla Stenberg) the information Don Luis wants and tells her it's her "passport"; he also gives her the address of her Uncle Emilio (Cliff Curtis), a criminal in Chicago, who will take care of her if she finds herself alone. The last thing he gives her is something that will keep her safe: his mom's necklace, the cataleya orchid. Fabio tells her to sit at the kitchen table and not move no matter what. After saying their goodbyes, Fabio and his wife Alicia (Cynthia Addai-Robinson) leave to battle Marco and his men but both of them are gunned down as Cataleya listens from the kitchen. Marco tries to manipulate her into giving the information. When he asks what she wants, she stabs him in the hand with a kitchen knife, saying "To kill Don Luis", and escapes. She safely makes it to the U.S. Embassy and gives the Embassy the information in exchange for a passport and passage to the United States. She escapes to the airport, where she makes it to the US and takes a bus to Chicago. Once she finds Emilio, Cataleya asks him to train her as an assassin.

A ship arrives, carrying toxic chemicals that have killed two of its crew members. Frank Martin and Inspector Tarconi fish in the Marseilles when Tarconi receives a call about a black Audi A8 that speeds past French customs and evades the police in a car chase. While Tarconi returns to headquarters to give Frank's alibi, in Odessa, Ukraine, the Environmental Agency Minister (Leonid Tomilenko) receives a threat from Ecocorp boss Johnson to reopen business negotiations. At night, the Audi crashes into Frank's home, with a wounded transporter, Malcolm Melville, whom Frank had referred when he declined a previous job. As paramedics take away Malcolm, Frank discovers in the backseat a woman who warns him not to take her from the car; she shows him a metal bracelet she and Malcolm have been wearing. Frank rushes outside, but the ambulance explodes. He is knocked out by a henchman.

Set in Mexico during the 1850s, the film tells the story of Sara Sandoval (Salma Hayek) and María Álvarez (Penélope Cruz). Frank Martin (Jason Statham) is a highly skilled driver known only as "The Transporter." He will transport anything, no questions asked, always on time, and he is known as the best in the business. He strictly follows three rules when transporting: Rule Number 1: "Never change the deal", Rule Number 2: "No names", and Rule Number 3: "Never open the package." Frank has been hired to transport some robbers from a bank heist. On delivery to their destination, they foist an extra man in the car so he refuses to drive until they decide to kill one robber. Liu Siu-jian (Jet Li), a Chinese intelligence agent, is sent to Paris to help apprehend Chinese mob boss Mr. Big (Ric Young), who is involved in heroin smuggling. He meets Inspector Jean-Pierre Richard (Tchéky Karyo), a corrupt and violent French police detective, at a hotel. Richard tricks Liu into believing he is simply providing reconnaissance of a meet involving Mr. Big. The plans are changed when Mr. Big is introduced to two prostitutes, one being Jessica Kamen (Bridget Fonda), an American woman, who takes him to his room to service him. While Liu and the rest are watching through the surveillance camera, Mr. Big kicks everyone out except for the two prostitutes. After pretending to seduce him, one of the prostitutes then stabs Mr. Big. Overseeing the events from a surveillance room, Liu rushes to stop the killing, but Richard enters shortly later to shoot Mr. Big and the prostitute with Liu's police-issued handgun, framing Liu for the murders.
