Masti revolves around three bachelors, Meet (Vivek Oberoi), Prem (Aftab Shivdasani) and Amar (Ritesh Deshmukh). Their lives are good until they get married and their lives turn to hell. Meet marries Anchal (Amrita Rao) who is obsessively possessive about her husband. Prem marries Geeta (Tara Sharma) who is overly religious and thus their sex life suffers. Amar marries Bindiya (Genelia D'Souza) who is dominating and lives with her mother (Archana Puran Singh). The friends get together one night and try to think of a way to get away from their wives for some time and relive their fun bachelor days. The movie starts with the death of a dreaded don Tony (Boman Irani). Then the movie focuses on Pyare (Fardeen Khan), who is blind and Mohan (Vivek Oberoi) who is deaf after they meet a severe accident (they were actors). Both are friends and seem to be happy with their disability but they crave the love of a girl who will love them for who they are. They eventually come across two sisters Preeti (Esha Deol) and Priya (Amrita Rao). The men fall in love with them and begin to dream of a life with them. Unfortunately, the girls have the opposite feeling and see them only as friends and when they ask for the reason why they don't love them, they learn it is because of their disabilities. And so, burdened by their disabilities the girls leave for Bangkok as they have a show there.

The story continues from the previous film. The four good-for-nothing friends Roy (Deshmukh), Adi (Warsi), Manav (Jaaferi) and Boman (Chaudhary), are still conning people for a living. One day, they see their enemy Kabir (Sanjay Dutt) driving the luxurious Mercedes Benz, and disguise themselves to find out the secret behind his success. They investigate and find that he lives off his wife's wealth. They then blackmail him into making them his business partners, but little do the four idiots know that Kabir and his wife Kamini (Sherawat) along with his sister Kiya (Ranaut) have an agenda of their own. Kabir connects them with an investor named Bata Bhai (Satish Kaushik) who is also a part-time don. The four friends steal Bata Bhai's money, and the money is then stolen by Kabir and his family. Kabir leaves the country along with his family, and leaves the four idiots to get killed by Bata Bhai. However, the four friends leave the city and manage to track Kabir down in Macau. They make a plan to ruin Kabir's happiness and take their money back by going in disguise. Roy cons Kiya by pretending to be in love with her, whilst Adi pretends to be Kabir's secretary. Boman poses as a woman in love with Kabir to break his relationship. Manav poses as various characters. Though, Kabir, Kamini, and Kiya have their own plan. They caught the four of them in their act.

Rana Mahendra Pratap Garewal (Mukesh Khanna) is a wealthy businessman along with his brother Vishwa (Dalip Tahil). They are good friends with Brijnath (Birju) (Paresh Rawal) who is initially very rich but loses all his wealth in a fire in his factory. The Garewal brothers had initially proposed the marriage of Rana's younger sister Madhu (Madhuri Dixit) and Brijnath's younger brother Raja (Sanjay Kapoor) (both are young; their brothers are adults). They break the marriage off. Shocked and saddened at the rejection by his best friends, Birju goes mad and gets electrocuted by accident. Raja then stays in the city outskirts with his brother taking care of him by doing petty jobs.
