After a mysterious gas attack which kills off most of the Earth's population, a few survivors gather at a country inn to figure out a plan for survival. However, the gas attack is only the first step in an alien invasion, in which groups of killer robots stalk the streets, able to kill anyone with the a mere touch of their hands. The group's members find additional weaponry in a nearby drill hall, but the robots continue their campaign of terror, which only increases when their victims rise from the dead as zombies, eager to kill anyone who might try to stop them. Yet despite frictions within the group -- and the birth of a baby, which further complicates matters -- most of the members survive. After discovering that the robots are being controlled from a local transmitting tower, the survivors blow it up and head to a nearby airport, where they commandeer a plane and fly towards an unknown destination, hoping additional survivors see their plane and join them.

Lee Barrett, a private investigator and former intelligence agent discharged for his outspoken views, is approached by a man with a tempting offer to join a political organization opposing bioweapons. His refusal proves the correct response, as the man is an impersonator sent by his former boss Eric Cavanaugh to test his loyalty. Barrett is asked by Cavanaugh to investigate the murder of the security chief of Station Three — a top-secret bioweapons laboratory station in the desert of Southern California — and the disappearance of its director and head scientist, Dr. Baxter. After they arrive at the station and wait for a time lock on the sealed lab to open, they are advised by another scientist, Dr. Gregor Hoffman, to seal the lab using concrete. Hoffman informs them that there are two lethal bioweapons in the lab, a strain of botulinus that oxidizes eight hours after its release, and a recently developed virus that he calls the "Satan Bug", which could kill all life on Earth in a matter of months. Determined to discover what happened in the room and taking extreme cautions, Barrett enters to find Dr. Baxter dead, with the vials containing the "Satan Bug" and 1200 grams of botulinus missing.

Set between the events of Dead Rising 2 and Dead Rising 3, The story follows online reporter Chase Carter and his camerawoman Jordan as they cover the stories of the people inside of the walled-in quarantined area in Oregon, as the government (running an organization called FEZA—Federal Emergency Zombie Authority) attempts to contain a viral outbreak that turns people into ravenous zombies. A television reporter named Angela Vidal (Jennifer Carpenter) and her cameraman Scott Percival (Steve Harris) are assigned to follow firefighters Jake (Jay Hernandez) and Fletcher (Johnathon Schaech) on their night shift. The crew responds to an emergency call from an apartment building. The apartment manager Yuri (Rade Sherbedgia) says that he and other residents heard screams from the room of an old woman named Mrs. Espinoza (Jeannie Epper), who has locked herself in her apartment. Jake, Fletcher, Yuri, police officers Danny (Columbus Short) and James (Andrew Fiscella), and the camera crew go to the apartment, where they find Mrs. Espinoza in serious condition. She is bleeding severely and foaming at the mouth. Moments later, they are attacked by Espinoza who bites James in the neck. They take him downstairs for medical assistance, while Fletcher stays with a now sedated Mrs. Espinoza upstairs. However, when they get downstairs, they find the apartment doors have been locked from the outside, leaving everyone, including several residents, trapped inside. Upstairs, Fletcher is attacked by Mrs. Espinoza and thrown from the railing to the ground, leaving him in serious condition. Lawrence (Greg Germann), a veterinarian, starts tending to the injured as best as possible.

The film begins in Leadville, Colorado. Trevor and his girlfriend, Nina, are in an abandoned building having a romantic encounter. They then head back to their respective parents' homes. They see a long line of cars at a military roadblock sealing off the city. An angry man yells at the officers because his child is sick and needs medical care. There has been an outbreak and medical help is needed throughout town. They are introduced to Captain Rhodes and Corporal Sarah Cross. Soon after, privates Bud Crain and Salazar show up and Bud immediately falls in love with Corporal Cross. Cross takes Bud to go with her while she visits her sick mother. When she arrives, Sarah talks to her brother (Trevor) and checks in on her mother. She then goes to see Trevor's friend, Kyle, after she learns his symptoms are similar to her mother's.
